Power Outages and ECIS Data Acquisition  

As we move through the summer season with occasional thunderstorms, the likelihood of a power
outage in the laboratory can be very real.  Should this happen while you are taking ECIS data, it is reassuring to know that the data acquired by your instrument are continually being stored on the PC's hard drive.  Thus, none of the data acquired before the outage are lost.  In addition, if the data are being gathered by a laptop that remains powered by its battery, when the power outage is over, the system will automatically resume taking data with a gap in the display over the time of the outage.
  

 

If your system is being controlled with a computer that has no battery backup, then with loss of power all data acquisition will be halted and it will be necessary to restart the system. If power is regained in a reasonable time, it is possible to rescue the experiment by following the instructions below: 

  • Reboot the ECIS software
  • Do the normal Setup procedure
  • Start a new time course file with exactly the same parameters used before the outage - the same acquisition program (for example MFT), the same frequencies, the same naming of the array (for example 8W10E+) and the identical selection of wells. 
  • Continue with the experiment and when all is complete finish data acquisition 

Now you have two files - one before and one after the power outage.   In the data analysis program these datasets can now be appended. 

 

Open the two files and display the first file; click on the Edit tab and then select <Append Data Sets> and choose the second file.  The display now shows the two files combined including the time gap between them when no data were collected.

 

In the example below, cells were inoculated in an 8W10E+ array at time zero and then approximately 1.25 hours into the run there was a loss of power for about 45 minutes.  When power returned, a new run was started and the two files were joined with the append software.  Notice that during the outage there are no data points, and the gap has been filled with a straight line for each well.

 

 

 

This new combined data set may now be explored with the analysis software and, if desired, stored as a .csv file.

 

Note:  this ability to append files is also handy should one inadvertently terminate data acquisition and want to continue an experiment with little interruption in the time course measurement.

Software Tip:

 

All ECIS arrays now come with a printed serial number and barcode for easy identification. When setting up an experiment this information can be used to specify the array type and track the array used in the comments. To do this automatically, select the menu item from Acquire | Array Serial # | Use Array Serial #. Then click Setup and enter the serial number when prompted. To manually enter or change the serial number select Enter Array Serial # from the same menu. The letter prefix specifies the array type, and the number is 6 or 7 digits.
 

A barcode scanner can also be used to scan the code and automatically populate the dialog box. One shortcut is that when using 2 arrays, you can scan the A and B codes as one entry and the software will interpret them as separate items.
 

An appropriate barcode scanner is available from Applied BioPhysics.

ECIS electrodes (8W4E-µ) in combination with 3T3 cells and Sonja Lukic's careful cell culture technique have created a funny face in one of the wells. We enjoyed the little fellow a lot. Obviously, he got involved in some fight considering the swelling on his head. Maybe he got wounded???
