Chenomx Profile2D - Assignment Confirmation Analysis Service
Chenomx's patented "Targeted Profiling" technique is a powerful tool that allows researchers to assign and quantify metabolites using 1D spectra.  Now, the confidence in assigning these metabolites can be enhanced by adding 2D analysis.  Chenomx is pleased to offer Profile2D: a new service aimed at confirming 1D metabolite assignments made using Chenomx Profiler, by 2D NMR analysis.

How it works:
Over the years, Chenomx has collected and curated a comprehensive 1H NMR metabolite library.  Chenomx has now developed this library further to include 2D 1H-1H NMR coupling patterns.  Together with our patented "Targeted Profiling" technique, the Profile2D service will increase confidence in the assignment of metabolites in complex biofluid mixtures, by using 2D cross peaks.

What we need:
It's easy! You can either send us these acquired spectra or we can use our in-house spectrometers to acquire them for you. For this service we will require a 1D spectrum using a standard 1D NOESY pulse sequence, and a 2D 1H-1H zdipsy-TOCSY spectrum using 256 increments, 32 transients, and a mixing time of 50 ms. 

What you get:
Increased confidence! The results of this analysis will be a 1D spectrum assigned using Chenomx NMR Suite, along with a list of metabolites that were assigned in 1D, as confirmed using the Profile2D service analysis.

Publications Using Chenomx NMR Suite 
http://chenomx.com/publications/index.htmlIn the May 2009 issue of Analytical Chemistry, there was a new study published by Um et al. entitled Pattern Recognition Analysis for the Prediction of Adverse Effects by Nonsteroidal Anti-Inflammatory Drugs Using 1H NMR-Based Metabolomics in Rats.  This paper used Chenomx NMR Suite to rapidly identify and quantify metabolites of interest using spectral binning and the patented "Targeted Profiling" technique. In this study, pattern recognition analysis of the 1H nuclear magnetic resonance (NMR) spectra of urine was performed to develop surrogate biomarkers related to the gastrointestinal (GI) damage induced by NSAIDs in rats. The results of global and targeted profilings suggest that the gastric damage induced by NSAIDs can be screened in the preclinical stage of drug development using a current metabolomics study. In addition, the putative biomarkers might also be useful for predicting the risk of adverse effects caused by NSAIDs.

Chenomx NMR Suite 5.1 Tips
Chenomx NMR Suite When using the Profiler module in Chenomx NMR Suite 5.1, compounds that have been modified in some way from their default state (including concentrations, transforms, custom colors, pins and stars) will appear in the Profiled Compounds set, and will appear in bold in the Compound Table.
Housekeeping
Please note, in our last newsletter (May 2009) we made a reference to certain metabolites that are only identifiable using NMR technology.  The listed metabolites were identified in a specific study comparing NMR to GC-MS and results are not necessarily the same in all studies and/or MS methods.
