In this issue of our newsletter we discuss our Quick Screen Project service and describe the different product formats available for AlbuPure® - our premium product for the purification of albumin-fusion proteins.  

 

We would also like to introduce the new 'Lunch & Learn Seminars' which provide customers with an opportunity to obtain one-to-one information about any of the bioseparation products or custom services available from ProMetic BioSciences Ltd (PBL).

NewsQuick Screen Projects 

 

PBL ScientistWith dedicated project managers working alongside scientific  experts within the fields of assay development, high throughput screening and downstream processing, PBL's Quick Screen service enables rapid identification of a chromatography adsorbent specific to your target protein. You provide the feedstock and process parameters and PBL will identify the most suitable PBL adsorbent and optimal conditions specific for your application - in just three steps:

  • STEP 1 - Rapid screening of PBL's range of existing chromatography adsorbents in PuraPlate™ format,
  • STEP 2 - Verification of lead candidates identified from library screening performed using packed columns and automated chromatography workstations,
  • STEP 3 - High throughput process optimization of chromatography parameters and buffers.
Once a lead candidate and optimum chromatography conditions have been identified from the Quick Screen Project, PBL will provide a comprehensive report describing the adsorbent(s) identified and optimized conditions for use. We will also provide a sample of the selected adsorbent.

AlbuPure® - Affinity Capture Adsorbent

 

Human serum albumin (HSA) has a vast array of applications within the biopharmaceutical industry including as a fusion partner to extend the half-life of a protein drug whilst maintaining it's activity. 

In order to support the rise in the production of a wide range of recombinant albumin-fusion proteins, an affinity matrix has been developed by PBL that is highly stable, robust, non-toxic and provides high binding capacity, recovery and purity - AlbuPure®

 

AlbuPure®, developed in collaboration with Novozymes and manufactured exclusively by PBL, is produced in a controlled environment to ISO 9001 quality standard and is designed for use in cGMP manufacturing of biological molecules. AlbuPure® is available in a variety of product formats and volumes from bench scale applications to the largest biomanufacturing processes.

AlbuPure® Product Formats:

  • ADSORBENT SLURRY - For column packing, from bench scale (25 mL - 1000 mL) to process scale (single batches of adsorbent up to 275 L);
  • Example Atoll ColumnsPRE-PACKED COLUMNS (PBL) - For attachment to chromatography workstations (1 mL column volume);
  • PURAPLATE™ - 96-column plate for rapid screening and purification method development (0.25 mL column volume);
  • ATOLL COLUMNS - AlbuPure® is available in ATOLL MediaScout® RoboColumn or MiniChrom columns to provide a platform for rapid process optimization (i.e. binding/loading, wash, elution and CIP applications).

PBL Downstream Processing & Applications Specialist

PBL has pioneered the design, development and manufacture of affinity separation products and technologies now used by major biopharmaceutical manufacturers worldwide. PBL's range of off-the-shelf products and custom designed chromatography adsorbents are complemented by comprehensive technical support.

 

PBL's Technical Support Team is a dedicated and experienced team focused on the provision of technical information and advice to users of our products. Members of the Technical Support Team are available by phone or by email to answer your technical questions. Our skilled team is able to provide training for all PBL products (on-site or by webcast presentations) as well as providing general information and advice on downstream process development.

A key member of PBL's Technical Support team is Buzz Lobbezoo. Buzz joined PBL in July 2004 as a research scientist in the downstream purification team and was recently appointed as the Downstream Processing & Application Specialist. In this role he works within the Technical Support team supporting PBL's products and customers. Mr. Lobbezoo has spent over 14 years in both the research and biotech sectors and has significant processing experience having worked on small-scale purification applications to large-scale manufacturing processes.   

  

The PBL Technical Support Team is always on hand to provide assistance with:

  • Q and A: The Technical Support Team has significant experience with all of PBL's products and is able to provide a quick response to customer questions.
  • Product literature: An expanding array of technical literature is produced by the Technical Support Team, from technical user guides to application notes and other associated literature to ensure that customers have all the information required for successful use of our products.
  • Application support: We can help with specific queries about your process such as ways of improving purity levels, increasing yields and reducing costs. The Technical Support Team is also able to undertake contract R&D to provide a specific solution for your processing needs.
  • Our Expertise: The Technical Support Team is available to provide assistance with tasks such as product evaluation and column packing. We have experience with purification processes from laboratory scale to 2 m diameter process columns.

(1) Question:  Dear Dev, what is the working conductivity range for your

new albumin fusion binding adsorbent AlbuPure®?

 

Answer:  With regards to AlbuPure® we have demonstrated high binding capacities, and subsequently high purity and recovery of the target material in combination with a series of increasing pH washes and elution with 10 mM sodium caprylate, when loading feedstock at low conductivity (3 mS/cm). Additional work by our Technical Support Team using yeast lysate containing albumin fusion protein demonstrated good results when loading with a conductivity of ~10 mS/cm (~pH 5.5).

The conductivity of the feed does have to be low to achieve high binding capacities and increasing the conductivity (and/or pH) of the load can affect binding which could result in a slight reduction in binding capacity.

 

(2) Question:  Dear Dev, I was wondering whether ProMetic BioSciences Ltd has any products which bind non human albumin?

 

Answer:  The short answer is YES, we do. Although we have not extensively tested our range of adsorbents with every type of animal serum albumin, our family of Albumin binding adsorbents, including Mimetic Blue® SA HL P6XL and AlbuPure®, will bind albumin from other sources including bovine serum albumin.  

 

(3) Question:  Dear Dev, we have adopted existing conditions for the purification of albumin fusion protein. Our target protein recoveries are excellent and our goal is to improve on host cell protein clearance while maintaining the recovery. Do you have any suggestions?  

 

Answer:  The AlbuPure® ligand was designed to bind to albumin fusion proteins with a high degree of selectivity. Consequently, the AlbuPure® column may be loaded using your existing equilibration and loading conditions. Once the column has been loaded with the target protein, it is important to optimize intermediate wash steps to ensure that non-specifically retained impurities (which can also be retained by the fact that a fully loaded column will act in similar manner to immobilized protein ligand, which may interact with host cell proteins) are removed before the elution step. Intermediate wash steps comprising of 50 mM sodium phosphate buffer, pH 6.0 and 50 mM sodium phosphate buffer, pH 7.0 are highly effective in removing non-specifically retained host cell proteins.
